The Trinity church stands in Kvemo Machkhaani, the lower of a pair of villages of the same name. The prefixes kvemo and zemo mean lower and upper and almost always work as a pair in Georgian toponymy.

The upper village, Zemo Machkhaani, lies about a kilometre to the north and higher up the slope; there stand the Armationi church of Saint George and a church of the Virgin. Such pairs formed over centuries as a settlement moved up or down a slope, and through them the history of how the land was settled can be read. Both Machkhaani villages lie south-east of Sighnaghi, the walled hilltop town.
